You definitely have the character and design of Twilight correct. Very well done!

I see two issues which you might want to address, one technical and one animation-specific.

The technical issue concerns outlines. During the "wince" portion (during the rainbow-zap) of the animation, the character outline on her face disappears. It seems like the outline might be ducking behind the fills on the head and the eye. Additionally, during the first portion of the animation (before any other action), it looks like the line for the back of Twilight's mane is too short; there is a gap where it meets her ear.

The animation issue is that the character's stance is fairly static; we're going from pose to pose then freezing within that pose. Just a little choppy. A little more anticipation and follow-through might be in order. One specific item to look at is the changes in iris-size for the eyes; I feel this could be used to greater effect going from key to key.

Nitpicks aside, this is Twilight. You've captured her very, very well.

The rigging is flawless as far as I can tell, but her motions are very stiff especially in the reaction and flowing action of the wincing part, have her at least looking up at some point after the wincing away from the ball shooting anyway from her before looking around to see if anyone else saw that bit of strangeness.

Don't be afraid to make her go big on her reactions to the ball going all wacky and flying away from her, for example make her jump back a little with her eyes going big before wincing away from the ball, also where Twilight winces her face becomes very stretched out and keep the stretched look.
